Usage : The quantitative determination Vitamin B-12 in circulation using human plasma or serum as sample.

Description With the advent and easy availability of vitamins and minerals determination of Vitamin B-12 in the circulation has gained considerable scrutiny. Vit B-12 is a water soluble vitamin and plays a significant role in the formation and equilibrium of red blood cells. It also helps form a protective sheath (myelin) around the nerves. Vitamin B-12 deficiency may take months to years to manifest depending on the cause and severity of the condition. Most common causes of Vit B-12 deficiency include diet and age. Useful tests to distinguish between Vit B-12 and Folate deficiency are methylmalonyl CoA (MMA) and homocysteine (hcy). Both manifest in similar symptoms; however even though both show increased levels of hcy only Vit B-12 deficiency causes increased levels of methylmalonyl CoA. Deficiency of Vit B-12 causes increased levels of hcy and MMA that are deemed to be responsible for oxidative stress that can cause coronary heart disease, atherosclerosis and neurodegenerative conditions like Parkinson‟s. Microplate enzyme immunoassay provides the technical advantage of optimal accuracy, precision and sensitivity with fewer manipulations. Extracted sample (patient serum or plasma, calibrators and controls) are added to designated wells. A specific antibody to human Vitamin B-12 labeled with biotin is added followed by an enzyme labelled folate solution. The enzyme labelled folate and endogenous folate compete for limited but specific sites on the antibody. After a predetermined incubation the microwells are washed to remove excess serum proteins and enzyme. A suitable substrate is added to generate inversely proportional color signal. From specified calibrator concentrations plotted on graph the Vitamin B-12 concentrations in the unknown samples can be interpolated. Advent of user friendly software and integrated readers, or even analyzers, has made calculations very easy for interpretation.
