Usage : The quantitative determination Alpha-fetoprotein (AFP) in circulation using human plasma or serum as sample.

 

Description Alpha-fetoprotein (AFP) is a single chain glycoprotein with a molecular weight of 70 kDa and 4% carbohydrate content. AFP is the major protein in fetal circulation and is closely related to albumin both genetically and structurally. It is one of the oncofetal antigens produced during fetal life and has a high concentration in fetal sera. This concentration decreases markedly or becomes undetectable after birth. It may be found in elevated levels in some malignancies such as hepatocellular carcinoma (HCC), trophoblastic disease and germ cell carcinoma. Even though there have been no randomized clinical trials showing a mortality risk benefit from the use of AFP as a screening tool for HCC some retrospective studies done in the east have shown improved survival with AFP screening. AFP has been advocated as a diagnostic tool for high risk populations in area like China, Africa, Japan and Alaska where the prevalence of HCC is high. An elevated level of AFP (> 10 µg/L) and an elevated serum Bilirubin (> 2.0 mg/dl) have been shown to be associated with shorter survival times. Elevated levels of AFP have also been associated with other types of cancers such as stomach, lung, colon, breast and lymphoma. Microplate enzyme immunoassay provides the technical advantage of optimal accuracy, precision and sensitivity with fewer manipulations. Sample (patient serum or plasma, calibrators and controls) are added to designated wells. A specific antibody to human AFP labeled with biotin and another specific antibody to AFP coupled with an enzyme are added to form a sandwich around serum AFP in the sample. After a predetermined incubation the microwells are washed to remove excess serum proteins and enzyme. A suitable substrate is added to generate directly proportional color signal. From specified calibrator concentrations plotted on graph the AFP concentrations in the unknown samples can be interpolated. Advent of user friendly software and integrated readers, or even analyzers, has made calculations very easy for interpretation.
